First post, but I've been coming down since the 90's. Here's my take:
Flight.
Flying into San Jose is the easiest and far better option than flying into Liberia. San Jose is a two hour +/- drive, while Liberia is a 4 hour +/- drive.
Getting to Jaco.
Bus: you can take the bus from the coca cola station. This is by far the cheapest option, but it also the most time consuming. Be careful at the bus station with your personal belongings because petty theft is common.
Taxi: There are one million taxi drivers waiting at the airport begging to take your bags from you and whisk you away. Whether you take one of these or prearrange a taxi before your arrival, you shouldn't spend more than $100.
Rental car: A great option for getting around but it can be a huge pain in the ass if you don't do your research ahead of time. Third party liability insurance is mandatory in Costa Rica. This means that extra $20 a day of insurance that is normally declined when traveling in the US or Europe, you are forced to take. Your first time renting a car you will feel like you are dealing with a "bait and switch" scam. Take lots of pictures of your car when you pick it up and document everything. You will also be obligated to put a pretty large hold on a credit card as a deposit for damages. Driving to Jaco is now very easy with the new highway.
Places to stay:
I favor the small vacation rentals over any of the hotels. Most of the year (besides major holidays) you can book once you get down there. You can arrive, get a lay of the land and then decide what you want. You can normally also save money by doing this because you can negotiate, especially if paying in USD. I'd stay away from the Marriott Los Suenos as there have been stories of the hotel giving the girls a hard time (they have the image to keep up).
Girls.
Many will recommend heading to the casino. There are always girls here, but prices tend to be much higher. I prefer heading to any of the popular bars in the area after sun down. There will always be a working girl and prices are better (R. I. P Beetle bar).

As for Cocal vs. Del Rey, first of all I hear the Del Rey is closed for renovations. Even so, all the best girls from there go to the Cocal anyway. With the new road from San Jose to Jaco it's only 90 minutes by car so the girls can work the Cocal by night and drive home.

Me and some friends are planning a trip to Jaco in February. Is it possible to rent from airbnb and bring back a girl from the Cocal Hotel without any problems? Since there is a group of us coming, I need to plan in case the Cocal is booked up. Airbnb seems like a cheap option, but will it work for getting the girls from the hotel or local bars? Also, is Jaco better than Hotel Del Rey in San Jose? I hear many say San Jose is the place, but others think Jaco has much better women. Any thoughts?

In Jaco, you don't need to depend on Cocal casino, in the night almost every bars and disco have working girls, these girls usually charge more than the usual price from Cocal (generally better quality than Cocal).
Most of the time I went to jaco with heredian chica (good friend for long now), and she will take me to all these places, sometime we wil pick another girl for us, sometimes just drink and party.
Even the same applies to Manuel Antonio, the only time which I felt was dry, is during Easter week.
