There are 20 different provinces there, and roughly a third of the population of Thailand live in that region. It is also, by any metric, the poorest part of Thailand. Agriculture is the largest sector of the economy so most local families are farmers, mainly growing rice. This relative poverty leads to the younger members of any family leaving to earn enough income to help support themselves and especially their family. For young ladies the best prospects are found in the entertainment / nightlife industry, mainly in the tourist areas of Bangkok, Pattaya, and to a lesser extent Phuket. This answers your first question. To answer your second question, there aren't such good prospects for finding ladies on a short trip to Isaan, at least nothing like the scene in Bangkok or Pattaya. There is actually a thread here in the Thailand section dedicated to the "Isaan Region" you could check out for a better idea.

Aside from their advice, I would add Siam Square, which is directly connected to Chulalongkorn University. At lunch times and starting in the afternoon, the girls will be instantly identifiable as uni girls by their uniforms. You will see them eating or browsing the shops. Siam One is a new (er) multi-story mall in Siam Square which caters to your target group.

You will probably need to employ a bit of "game", but should be able to get an idea of their interest in a foreigner fairly quickly with some small talk. From that, do your thing--get a phone number or line or whatever. Just see how it goes.

Lots of hotels in various price ranges nearby to stay at, but you can get to the Siam Center BTS station easily from most parts of the city.
